Improved radioenzymatic assay for spectinomycin.

The sensitivity and reproducibility of the radioenzymatic adenylylating assay for spectinomycin were improved by modifications of the assay procedure and by partial purification of the enzyme. The presence of Bacillus subtilis ribosomes or S-150 fraction in the enzymatic reaction mixture interfered with the ability to measure spectinomycin by this method.

Radioenzymatic assay of catecholamines in plasma after a preliminary solvent extraction compared with an analogous liquid-chromatographic method.

Catechol-borate complexation and ion-pair formation combined with organic solvent extraction as preliminaries to radioenzymatic assay (REA) with catechol-O-methyltransferase-catalyzed methylation allows the quantification of catecholamines in plasma samples of any volume. We attained improved detection limits for 1-mL samples: 9 ng/L for norepinephrine (NE), 8 ng/L for epinephrine (E), and 11 n...

Comparison of improved fluorometric methods used to quantitate plasma catecholamines.

Comparative studies reveal that the ethylenediamine and the trihydroxyindole method when employed as suggested are suitable for assessing sympatho-adrenal activation under various stressful conditions that are accompanied by release of catecholamines into the circulation. Both methods are very accurate in quantitating epinephrine or norepinephrine that has been added to plasma or whole blood, a...

Simplified radioenzymatic assay for chloramphenicol.

A simplified radioenzymatic assay for chloramphenicol was developed by eliminating the need for cumbersome extraction procedures. After the acetylation of chloramphenicol with [(14)C]acetyl coenzyme A in the presence of chloramphenicol acetyltransferase, the reaction mixture was added to a toluene-based scintillation fluid. Since (14)C-acetylated chloramphenicol is more soluble than [(14)C]acet...

Direct determination of angiotensin-converting enzyme inhibitors in plasma by radioenzymatic assay.

We describe a simple, rapid, specific radioenzymatic assay for "CGS 16617," a new, potent inhibitor of angiotensin-converting enzyme (ACE; EC 3.4.15.1) in human plasma. This assay is based on the principle that the inhibitor (i.e., the drug) binds to the ACE in plasma and hence the amount of free ACE in plasma is inversely related to the amount of active inhibitor present. Free enzyme is reacte...
